Alright, so I've got quite a bit of yellow nutsedge coming in throughout my yard. I'm only going to spray a small section, probably 1000 to 2000 sq ft, but for the sake of math lets just say 1000 sq ft. I just want to make sure that I'm reading this right, but I'd need to apply at the 1.25 oz rate to kill the yellow nutsedge. Would that be 2.5 small scoops to the 1 gallon of water? Also, I could decide to spot treat those as well, and probably get away with a half gallon. That could be an option as well.

I was going to provide a quick answer, but then I reread the label and this is actually a great question. The application dosage is 1.25 which equates to 5 scoops, but the delivery method is 1 gallon of water instead of 2. So either 5 in 2 gallons or two 2.5 in 1 gallon mixes to cover 1000 sq ft.

Now my question is how importnat is the 2 gallon requirement if a sprayer is calibrated for 1 gallon every 1000 sq ft?

yeah, I'll probably mix that with a little Celsius as well to kill any other nasties trying to come up. Has anyone had an issue with using the marker dye with certainty? It states that it may not be as effective if used

It's not! same concentration...and you already give the condition that the sprayer is calibrated so you would be getting the correct amount down per unit surface area.
